چکیده

The subchronic safety of sanguiritrin, a mixture of sanguinarine (SA) and chelerythrine (CHE) quaternary benzo[c]phenanthridine alkaloids (QBA), obtained from Macleaya cordata was assessed. Rats were fed a diet containing 120 ppm sanguiritrin (100 ppm QBA) for 109 days. The feed consumption and the animal weight were monitored. The content of QBA in selected tissues and plasma was determined using HPLC. It was evidenced that 2% of QBA were absorbed through the GIT while 98% were excreted in the feces. In plasma, bilirubin, urea, creatinine, glomerular filtration, AST, ALT, GMT, ALP and total antioxidant capacity were determined. In liver, GSH level, lipoperoxidation products, SOD and GPx activities and total amount of cytochrome P450 were evaluated. Damage to nuclear DNA was assessed; a 32P-postlabeling assay proved that no DNA-adducts were detected in nuclear and mitochrondrial DNA in liver. No adverse effects were observed on rat organism. QBA had no influence on the gut mucosal epithelium, liver tissue and any biochemical parameters tested. Oxidative stress was not manifested during the experiment.

Integration of Transcriptome, Proteome and Metabolism Data Reveals the Alkaloids Biosynthesis in Macleaya cordata and Macleaya microcarpa

BACKGROUND The Macleaya spp., including Macleaya cordata and Macleaya microcarpa, are traditional anti-virus, inflammation eliminating, and insecticide herb medicines for their isoquinoline alkaloids. They are also known as the basis of the popular natural animal food addictive in Europe. However, few studies especially at genomics level were conducted on them. Hence, we performed the Macleaya ...

Macleaya cordata extract and Sangrovit genotoxicity. Assessment in vivo.

BACKGROUND Sanguinarine (SG) has been reported to form DNA adducts in vitro and increase the levels of DNA single strand breaks in the blood and bone marrow of mice treated intraperitoneally with SG. Recently, we showed no genotoxic effects of orally administrated 120 mg/kg feed Macleaya cordata extract (a mixture of sanguinarine and chelerythrine) in pigs or rats in 90-day studies. The goal of...

Macleaya cordata Extract Decreased Diarrhea Score and Enhanced Intestinal Barrier Function in Growing Piglets

Macleaya cordata extract is of great scientific and practical interest to researchers, due to its antimicrobial and anti-inflammatory responses within experimental animals. This study was designed to determine the diarrhea score and innate immunity of growing piglets after they had received Macleaya cordata extract supplements. A total of 240 growing pigs were randomly assigned to one of three ...

Seasonal variation of bioactive alkaloid contents in Macleaya microcarpa (Maxim.) Fedde.

Macleaya microcarpa (Maxim.) Fedde belongs to the genus Macleaya, family Papaveraceae. Together with the better known and more frequently studied species M. cordata (Willd.) R. Br. it is a main source of quaternary benzo[c]phenanthridine alkaloids. Using HPLC we determined the content of eight isoquinoline alkaloids in the aerial and underground parts of 1-, 2-, 12- and 13-year old plants and f...

Analysis of the Impact of Isoquinoline Alkaloids, Derived from Macleaya cordata Extract, on the Development and Innate Immune Response in Swine and Poultry

Medicinal extract has been chronicled extensively in traditional Chinese medicine. Isoquinoline alkaloids, extract of Macleaya cordata (Willd.) R. Br., have been used as feed additive in both swine and poultry. Dietary supplementation with isoquinoline alkaloids increases feed intake and weight gain. In addition, recent researches have demonstrated that isoquinoline alkaloids can regulate metab...
